I don't have this model of hard drive or Syno (DS1511+), but in principle it should accept all USB hard drives. I have connected both 3.5" and 2.5" external drives, USB2.0 as well as USB3.0, without any problems.

Via the USB 3 port of my DS918+, I centralize all my backups on the Seagate without any problem. I think this is also applicable to the DS216play.
