Is it possible to clone from a hard disk to an external hard disk (via this USB 3.0 port on the back)? Thanks a lot

No. The box is merely a USB 3.0 to SATA module. The connector on the back is used to connect to a computer.

No, the cloning function is only for "naked" hard drives, if you have one with a USB connection, you have to do it manually via Windows Explorer.
